The replacement of the mouse Lag3 gene with its human ortholog enables the preclinical validation of anti-hLAG-3 compounds. Combined treatment with candidate anti-hLAG-3 compounds and Atezolizumab resulted in efficient tumor growth inhibition.

In vivo response of MC38 tumor-bearing humanized hLAG3 mice to combo-therapy.
Homozygous hLAG3 mice (n=8 per group) were inoculated with MC38 colon cancer cells. Randomization and treatment initiation were performed once the tumors grew to 70-80 mm3. A significant anti-tumor effect was observed when the antibody targeting human LAG3 was administered together with Tecentriq.
