The B6 hPD-L1 mouse features the replacement of the mouse B7-H1 gene with its human ortholog. This model has been shown to effectively predict the anti-tumor effects of candidate anti-hPD-L1 blocking antibodies.
A. Immunophenotyping comparison of the spleens of homozygous hPD-L1 and WT mice.
B. In vivo response of MC38 tumor-bearing humanized PD-L1 mice to anti-hPD-L1.
Homozygous hPD-L1 mice (n=10) were inoculated with MC38 colon cancer cells. Randomization and treatment initiation were performed once the tumors grew to 100 mm3. Candidate compounds induced a significant tumor growth inhibition (p < 0.001).
